In the present investigation milt oí puye (Galaxias maculatus) was studied qualitatively and quantitatively. Adult specimens (6.24 ± 0.84 cm of standard length) were captured from fresh waters oí the South oí Chile and were kept in captivity. Once they were mature, the specimens were anesthetized and a sample of milt was extracted by stripping. This was subjected to visually examined and spermatic count, spermatocrit and sperm motility determined. The results obtained show that milt of puye is not very abundant (máximum 0.2 mi per individual), of a whitish color and great viscosity. Quantitatively, it was found an average of 55.3 x 106 sperm/ml, a spermatocrit of 89.63 % average with a correlation between both variables of r = 0.97. Motility check withdifferentactivatorsis veryslowand it wasonly observed a motility equal to 2 (in a scale of 0 to 5) for approximately 30 min when carbamide was used.
